目的:研究玉屏风散对小鼠腹腔巨噬细胞一氧化氮(NO)生成和诱导型一氧化氮合酶(iNOS)表达的影响。方法:制备玉屏风散水煎液,分离健康昆明种小鼠原代腹腔巨噬细胞进行体外培养,培养基中分别加入不同剂量上述药液使终末浓度分别达到1.67、3.33、8.33、16.67 g/L(折合生药)进行细胞培养,并设空白对照组。培养24h后,分别用Griess法和免疫细胞化学法检测NO生成量和iNOS表达量。结果:与空白对照组相比,玉屏风散加药组NO生成量和iNOS表达量显著增加(P<0.05)。结论:玉屏风散可以刺激小鼠腹腔巨噬细胞NO的生成,且可能通过诱导巨噬细胞表达iNOS,继而产生NO。这可能是玉屏风散增强机体免疫功能、抑菌、抗病毒等作用的途径之一。
Objective: To study the effects of Yupingfengsan on nitric oxide (NO) production and iNOS expression in murine peritoneal macrophages. Methods: The Yuping Fengsan Decoction was prepared and the primary peritoneal macrophages from healthy Kunming mice were isolated and cultured in vitro. Different doses of the above liquid were added into the culture medium to make the final concentrations reach 1.67, 3.33, 8.33 and 16.67 g / L (equivalent to crude drugs) for cell culture, and set the blank control group. After cultured for 24h, the production of NO and the expression of iNOS were detected by Griess method and immunocytochemistry respectively. Results: Compared with the blank control group, NO production and iNOS expression in Yupingfeng powder group increased significantly (P <0.05). Conclusion: Yupingfeng powder can stimulate NO production in mouse peritoneal macrophages and induce NO production by inducing iNOS expression in macrophages. This may be one of the ways Yupingfeng San enhance the body’s immune function, antibacterial, antiviral and other effects.
