目的研究葛根素注射液对Beagle犬红细胞的溶血作用。方法取24只Beagle犬的红细胞,分别与不同浓度的葛根素注射液(1~10 mg/mL)、丙二醇溶液(2%~20%)共同孵育,观察24 h内红细胞的状态和溶血发生率,用χ2检验统计分析溶血实验结果。结果增大葛根素注射液浓度、延长与供试品(红细胞)反应时间(葛根素10 mg/mL,丙二醇浓度20%,反应时间24 h),24只Beagle犬红细胞的溶血发生率高达66.7%,而等浓度丙二醇对照溶液组的溶血发生率为16.7%,两者差异有显著性(P<0.01)。结论与常规溶血试验方法相比,本研究中改进的溶血反应评价方法能够更准确检测出葛根素注射液所致的偶发性溶血反应,所致溶血反应主要由高浓度的葛根素所引起。
Objective To study the hemolysis effect of puerarin injection on Beagle dog erythrocytes. Methods Erythrocytes of 24 Beagle dogs were incubated with puerarin (1-10 mg / mL) and propanediol (2% -20%) respectively. Conclusion Compared with the conventional hemolysis test, the improved hemolytic reaction evaluation method in this study can detect the occasional hemolytic reaction caused by puerarin injection more accurately. The hemolysis is mainly caused by the high concentration of puerarin.
