目的:探讨槲皮素对HL鄄60细胞的形态、血管内皮生长因子(VEGF)表达的影响。方法:以HL鄄60细胞为研究对象,细胞形态学观察采用Wright染色法;膜联蛋白V(AnnexinⅤ)标记检测细胞凋亡率;VEGF表达采用ELISA法。结果:①经槲皮素处理后,HL鄄60细胞形态学上出现凋亡特征性改变。②槲皮素处理后HL鄄60细胞凋亡率明显增加。③槲皮素处理后HL鄄60细胞显著降低VEGF表达。结论:槲皮素在体外能抑制白血病细胞HL鄄60生长并诱导其凋亡;槲皮素可抑制白血病细胞分泌VEGF。
Objective: To investigate the effect of quercetin on the morphology of HL-60 cells and the expression of vascular endothelial growth factor (VEGF). Methods: The HL-60 cells were used as the research objects. Wright staining was used to observe the morphological changes of the cells. The apoptosis rate was detected by Annexin V and the expression of VEGF was detected by ELISA. Results: ① The morphology of HL-60 cells changed markedly after quercetin treatment. ② The apoptosis rate of HL-60 cells increased obviously after quercetin treatment. ③ HL-60 cells treated with quercetin significantly reduced VEGF expression. CONCLUSION: Quercetin can inhibit HL-60 cell growth and induce apoptosis of leukemia cells in vitro. Quercetin can inhibit leukemia cells to secrete VEGF.
