目的检测人食管癌组织中结缔组织生长因子(CTGF)基因mRNA和蛋白表达水平,了解食管癌患者术后生存情况及其影响因素。方法运用实时定量PCR技术测定186例食管癌患者的癌组织及配对的癌旁组织中CT-GF基因mRNA和蛋白的表达水平,采用Kaplan-Meier单因素与Cox多元回归进行生存分析。结果食管癌组织中CTGFmRNA表达水平(5.29±5.12)明显高于其配对的癌旁组织(8.63±5.12)(t=10.112,P=0.0000);食管癌组织CTGF蛋白表达水平(20.83±8.20)明显高于其配对的癌旁组织(4.52±4.53)(t=23.820,P=0.0000);Kaplan-Meier法生存分析结果显示,CTGFmRNA及CTGF蛋白的表达水平、性别、家族史、浸润深度、肿瘤大小、淋巴结转移、分化程度与患者的生存率明显相关;Cox回归分析结果显示,CTGFmRNA的表达水平、CTGF蛋白的表达水平、性别、家族史、淋巴结转移、分化程度是影响食管癌患者生存率的独立危险因素。结论食管癌组织中CTGF的高表达可能影响食管癌患者术后的生存,其可作为食管癌患者预后的一个评价指标,并可能成为基因治疗的一个作用靶点。
Objective To detect the mRNA and protein expression of connective tissue growth factor (CTGF) in human esophageal cancer and to investigate the postoperative survival and its influencing factors in esophageal cancer patients. Methods Real-time quantitative PCR was used to detect the mRNA and protein expression of CT-GF in 186 cases of esophageal cancer and paired paracancerous tissues. Survival analysis was performed by Kaplan-Meier single factor and Cox multiple regression. Results The expression of CTGF mRNA in esophageal cancer tissues (5.29 ± 5.12) was significantly higher than that in paired paracancer tissues (8.63 ± 5.12) (t = 10.112, P = 0.0000). The expression of CTGF protein in esophageal cancer tissues was significantly (20.83 ± 8.20) (4.52 ± 4.53) (t = 23.820, P = 0.0000). Kaplan-Meier survival analysis showed that the expression of CTGF mRNA and CTGF protein, gender, family history, depth of invasion, tumor size , Lymph node metastasis and differentiation were significantly correlated with the survival rate of patients; Cox regression analysis showed that CTGF mRNA expression, CTGF protein expression, gender, family history, lymph node metastasis and differentiation were the independent survival rates of patients with esophageal cancer Risk factors. Conclusion The high expression of CTGF in esophageal carcinoma may affect the postoperative survival of patients with esophageal cancer, which can be used as a prognostic indicator of esophageal cancer and may be a target of gene therapy.
