目的:探讨热休克蛋白70(Hsp70)、葡萄糖调节蛋白94(Grp94)、IgG在人肺癌组织中的表达及意义。方法:应用免疫组织化学方法结合病理学图像分析系统研究40例肺癌组织中Hsp70、Grp94和IgG的表达。并用免疫荧光双标染色及激光共聚焦技术研究三者的定位关系。结果:人肺癌组织中Hsp70、Grp94、IgG都有高表达,阳性率分别为65%(26/40)、45%(18/40)和82.5%(33/40),Hsp70阳性定位胞核胞质,Grp94和IgG阳性定位主要在胞质,平均光密度值分别为(5.10±0.32)、(3.52±0.35)和(8.12±0.31)。26例Hsp70阳性的肺癌病例中有10例与IgG有共定位关系,18例Grp94与IgG有共定位关系。结论:Hsp70、Grp94和IgG在肺癌组织中高表达,提示三者在肺癌的发生发展中起重要作用;肺癌肿瘤细胞表达的IgG与Hsp70、Grp94分别存在一定共定位关系,其在肿瘤的发展中可能具有协同作用或者在肺癌组织中存在Hsp70-IgG、Grp94-IgG和Hsp70-Grp94-IgG复合物的形式。
Objective: To investigate the expression and significance of heat shock protein 70 (Hsp70), glucose regulatory protein 94 (Grp94) and IgG in human lung cancer. Methods: The expression of Hsp70, Grp94 and IgG in 40 cases of lung cancer was studied by immunohistochemistry and pathological image analysis system. Immunofluorescence double-stained staining and confocal laser scanning microscopy were used to study the localization of the three. Results: The positive rates of Hsp70, Grp94 and IgG in human lung cancer were all 65% (26/40), 45% (18/40) and 82.5% (33/40) The positive localization of Grp94 and IgG was mainly in the cytoplasm with average optical density values of (5.10 ± 0.32), (3.52 ± 0.35) and (8.12 ± 0.31), respectively. Of the 26 Hsp70-positive lung cancer cases, 10 had colocalization with IgG and 18 had Grp94-IgG colocalization. CONCLUSION: Hsp70, Grp94 and IgG are highly expressed in lung cancer tissues, suggesting that they play an important role in the development of lung cancer. IgG and Hsp70 and Grp94 expressed in lung cancer cells may have a colocalization relationship with each other, which may play an important role in tumor development Has a synergistic effect or exists in the form of Hsp70-IgG, Grp94-IgG and Hsp70-Grp94-IgG complexes in lung cancer tissues.
