目的探讨结核分枝杆菌CFP10-ESAT6融合蛋白对不同分枝杆菌致敏动物的鉴别作用。方法以不同分枝杆菌分别致敏豚鼠,致敏成功后,每只豚鼠皮内分别注射结核分枝杆菌CFP10-ESAT6融合蛋白和结核菌素纯蛋白衍生物(PPD)。用双盲法记录注射后24和48h注射局部硬结的纵径和横径,计算其均值。结果PPD对结核分枝杆菌活菌、死菌和卡介苗致敏豚鼠的皮肤反应均为阳性,局部硬结直径分别为(13.91±1.04)mm、(13.11±1.40)mm和(13.16±1.43)mm。结核分枝杆菌CFP10-ESAT6融合蛋白对结核分枝杆菌活菌致敏豚鼠的皮肤反应为强阳性(≥10mm),局部硬结直径为(10.53±2.66)mm;对结核分枝杆菌死菌致敏豚鼠的皮肤反应为弱阳性(≤5mm)或阴性,局部硬结直径为(1.61±1.39)mm;对卡介苗致敏豚鼠的皮肤反应为阴性,局部硬结直径为0。结论结核分枝杆菌CFP10-ESAT6融合蛋白可鉴别结核分枝杆菌活菌、死菌和卡介苗致敏动物。
Objective To investigate the differential role of Mycobacterium tuberculosis CFP10-ESAT6 fusion protein in different mycobacterium sensitized animals. Methods The guinea pigs were sensitized with different mycobacteria. After sensitization, the guinea pigs were injected intradermally with M. tuberculosis CFP10-ESAT6 fusion protein and tuberculin pure protein derivative (PPD) respectively. Double-blind method was used to record the longitudinal and transverse diameters of local induration at 24 and 48 h after injection. Results The skin reaction of PPD was positive for the Mycobacterium tuberculosis, dead and BCG-sensitized guinea pigs. The diameter of the local lesion was (13.91 ± 1.04) mm, (13.11 ± 1.40) mm and (13.16 ± 1.43) mm, respectively. Mycobacterium tuberculosis CFP10-ESAT6 fusion protein had a strong positive (≥10mm) skin reaction to Mycobacterium tuberculosis-sensitized guinea pigs, and a diameter of (10.53 ± 2.66) mm for mycobacterium tuberculosis sensitized to Mycobacterium tuberculosis Skin reactions in guinea pigs were weakly positive (≤5mm) or negative, and the diameter of the local induration was (1.61 ± 1.39) mm. The cutaneous reaction to BCG-sensitized guinea pigs was negative and the diameter of the local induration was zero. Conclusion Mycobacterium tuberculosis CFP10-ESAT6 fusion protein can identify Mycobacterium tuberculosis viable, dead and BCG-sensitized animals.
