目的检测骨肉瘤组织中survivin及肿瘤坏死因子α(TNF-α)的表达,探讨其相关性。方法应用免疫组织化学二步法及RT-PCR法对62例骨肉瘤组织进行survivin、TNF-α的检测和分析,并与62例正常骨组织进行对照研究。结果骨肉瘤组中survivin及TNF-α表达均高于正常结膜组(P<0.05),Survivin和TNF-α的阳性表达与性别、年龄、肿瘤大小和组织学类型无关(P>0.05),Survivin和TNF-α的阳性表达与肿瘤的转移和生存情况密切相关(P<0.05),TNF-α与Sur-vivin蛋白表达显著相关(P<0.05)。结论骨肉瘤组织中TNF-α、Survivin mRNA的过度表达,提示骨肉瘤组织的发生与细胞凋亡抑制有关。
Objective To detect the expression of survivin and tumor necrosis factor-α (TNF-α) in osteosarcoma and to investigate their correlation. Methods The expression of survivin and TNF-α in 62 cases of osteosarcoma were detected by two-step immunohistochemistry and RT-PCR, and compared with 62 cases of normal bone tissue. Results The expression of Survivin and TNF-α in osteosarcoma group was significantly higher than that in normal conjunctiva group (P <0.05). The positive expression of Survivin and TNF-α was not related to gender, age, tumor size and histological type (P> 0.05) (P <0.05). The positive expression of TNF-α and Survivin protein were significantly correlated with tumor metastasis and survival (P <0.05). Conclusion Overexpression of TNF-α and Survivin mRNA in osteosarcoma suggests that the occurrence of osteosarcoma is related to the inhibition of apoptosis.
