目的观察高尿酸血症(HUA)大鼠血尿酸(SUA)与IL-6、转化生长因子β1(TGF-β1)的关系,以及非布司他对HUA大鼠血清IL-6和TGF-β1的影响。方法 SD大鼠随机分为对照组(Con)、模型组和非布司他干预组,每组12只,分别于第0、4、8周末检测大鼠24 h尿蛋白(24 hU;Alb),SUA、血肌酐(Scr)、BUN、IL-6和TGF-β1。采用HE及Masson染色法观察肾脏病理学改变。结果模型组及非布司他干预组SUA、Scr、BUN、IL-6和TGF-β1高于Con组(P<0.05),模型组IL-6和TGF-β1较非布司他干预组升高(P<0.05或P<0.01)。相关分析显示,血清IL-6与SUA、TGF-β1、Scr及24 hUAlb呈正相关(r=0.908、0.827、0.848、0.855,P<0.01);非布司他治疗能降低SUA、IL-6及TGF-β1水平(P<0.05)。肾脏病理学改变以模型组损伤最明显,非布司他治疗后肾脏损伤减轻。结论非布司他对HUA有明显的治疗作用,在降尿酸的同时还可降低IL-6及TGF-β1,减轻肾脏炎症反应,对肾脏功能和形态的损伤有很好的保护作用。
Objective To observe the relationship between serum uric acid (SUA), IL-6 and transforming growth factor-β1 (TGF-β1) in hyperuricemia rats and the effect of febuxostat on serum IL-6 and TGF-β1 Impact. Methods SD rats were randomly divided into control group (Con), model group and febuxostat group, with 12 rats in each group. 24 h urinary protein (24 hU; Alb) , SUA, serum creatinine (Scr), BUN, IL-6 and TGF-β1. The renal pathological changes were observed by HE and Masson staining. Results The levels of SUA, Scr, BUN, IL-6 and TGF-β1 in model group and febuxostat group were significantly higher than those in Con group (P <0.05). IL-6 and TGF- High (P <0.05 or P <0.01). Correlation analysis showed that serum IL-6 was positively correlated with SUA, TGF-β1, Scr and 24 hUAlb (r = 0.908,0.827,0.848,0.855, P <0.01) .Bupivat treatment could reduce SUA, IL-6 and TGF-β1 levels (P <0.05). Renal pathological changes in the model group the most obvious injury, febrileus lesion after the reduction of febuxostat. Conclusions Fefebuterol has a significant therapeutic effect on HUA. It can reduce the levels of IL-6 and TGF-β1 while reducing uric acid, relieve the inflammatory reaction of kidney and protect the renal function and morphology.
