目的 研究幽门螺杆菌 (Hp)源抗菌肽 (2 2 0 )对胃黏膜上皮细胞及胃癌细胞生长的影响。方法 采用琼脂扩散法检测Hp抗菌肽 (2 2 0 )及杀菌肽 (cecropin)B的抗菌活性及致死浓度 (LC) ,MTT法测定Hp抗菌肽 (2 2 0 )、杀菌肽B对胃黏膜上皮细胞株GES 1、胃癌细胞株SGC 790 1的细胞毒作用及半数抑制浓度 (IC50 )。结果 Hp抗菌肽 (2 2 0 )及杀菌肽B对E .coliK12D31的LC分别为 6 5和 0 .4 4μmol/L ,Hp抗菌肽 (2 2 0 )的抗菌活性较杀菌肽B低。Hp抗菌肽 (2 2 0 )及杀菌肽B对胃癌细胞具有浓度依赖性细胞毒作用。而Hp抗菌肽 (2 2 0 )在相对较低浓度时对胃黏膜上皮细胞有促进增殖的作用。杀菌肽B在 5~ 2 0 μmol/L浓度范围内对胃黏膜上皮细胞株GES 1无细胞毒作用。Hp抗菌肽 (2 2 0 )对胃黏膜上皮细胞株GES 1、胃癌细胞株SGC 790 1的IC50 为 6 30、5 0 0 μmol/L ,杀菌肽B对胃黏膜上皮细胞株GES 1、胃癌细胞株SGC 790 1的IC50 为 75、2 5 μmol/L。 结论 Hp抗菌肽 (2 2 0 )的抗菌活性较杀菌肽B低 ,对胃癌细胞具有浓度依赖性细胞毒作用。Hp抗菌肽 (2 2 0 )在相对较低浓度对胃黏膜上皮细胞有促进增殖的作用 ,可能与胃癌发生有关。
Objective To study the effect of Hp antimicrobial peptide (220) on the growth of gastric mucosal epithelial cells and gastric cancer cells. Methods The antibacterial activity and lethal concentration (LC) of Hp antimicrobial peptide (220) and cecropin B were determined by agar diffusion method. The inhibitory effect of Hp antimicrobial peptide (220) and cecropin B on gastric mucosal epithelium Cell line GES 1, gastric cancer cell line SGC 790 1 cytotoxicity and half inhibitory concentration (IC50). Results The LCs of Hp antimicrobial peptide (220) and bactericidal peptide B against E.coli K12D31 were 65 and 0.44 μmol / L, respectively. The antibacterial activity of Hp antimicrobial peptide (220) was lower than that of cecropin B. Hp antimicrobial peptide (220) and cecropin B have a concentration-dependent cytotoxic effect on gastric cancer cells. Hp antimicrobial peptides (220) at a relatively low concentration of gastric mucosal epithelial cells to promote proliferation. Cecropin B had no cytotoxic effect on gastric mucosal epithelial cell line GES 1 in the concentration range of 5 ~ 20 μmol / L. The IC50 of Hp antimicrobial peptide (220) on gastric mucosal epithelial cell line GES 1 and gastric cancer cell line SGC 790 1 was 6 30 and 50 μmol / L, and cecropin B had an inhibitory effect on gastric mucosal epithelial cell line GES 1, gastric cancer cells The IC50 of strain SGC 790 1 was 75,25 μmol / L. Conclusion The antibacterial activity of Hp antimicrobial peptide (220) is lower than that of cecropin B and has a concentration-dependent cytotoxic effect on gastric cancer cells. Hp antimicrobial peptide (220) at a relatively low concentration of gastric mucosal epithelial cells to promote proliferation, may be related to gastric cancer.
