本文应用肾小球细胞体外培养方法和双同位素标记技术,观察了阿霉素肾病大鼠肾小球系膜细胞(GMC)对白细胞介素6的反应、肾病大鼠GMC条件培养液(GMC-CM)对正常大鼠GEC掺人~(35)S和~3H-亮氨酸的影响以及GMC-CM对小鼠骨髓瘤杂交瘤细胞增殖的作用。结果显示:①肾病大鼠GMC对不同浓度IL-6的反应曲线与正常GMC明显不同;②肾病极期GMC-CM明显促进正常大鼠GEC合成含硫化合物;③肾病大鼠GMC-CM中含有骨髓瘤细胞的杀伤物质。以上结果提示,阿霉素肾病大鼠GMC的部分生物活性发生了改变。
In this study, glomerular cells cultured in vitro and double isotope labeling technology, observed adriamycin nephropathy rat mesangial cells (GMC) on interleukin 6 response, nephrotic rat GMC conditioned medium (GMC- CM) on the incorporation of ~ (35) S and ~ 3H-leucine into normal rat GEC and the effect of GMC-CM on the proliferation of mouse myeloma hybridoma cells. The results showed that: ①The GMC-CM of nephrotic rats significantly differed from that of normal GMCs in response to different concentrations of IL-6; ② GMC-CM significantly promoted the synthesis of sulfur-containing compounds in normal rats; Myeloma cells kill substances. The above results suggest that part of the biological activity of GMC in adriamycin-induced nephropathy rats has changed.
