目的:优选首乌藤改善睡眠有效部位的微波辅助提取工艺。方法:采用单因素试验考察微波功率、提取时间、乙醇体积分数、溶剂用量、提取温度等对首乌藤总黄酮和总蒽醌含量的影响。在单因素试验基础上,选取微波功率、提取时间、乙醇体积分数为考察因素,每个因素设置5个水平,以总黄酮、总蒽醌含量的总评“归一值”为评价指标,利用星点设计-效应面法优选微波提取工艺。结果:首乌藤改善睡眠有效部位的最佳微波提取工艺为微波功率585 W,提取数1次,加10倍量82%乙醇于60℃提取11 min。结论:优选的微波辅助提取工艺简便易行,预测性良好,为首乌藤后续的剂型开发和工业化生产提供实验依据。
OBJECTIVE: To optimize microwave-assisted extraction of the effective parts of Morus alba in sleep improvement. Methods: The effects of microwave power, extraction time, ethanol volume fraction, solvent dosage and extraction temperature on the content of total flavonoids and total anthraquinones were studied by single factor experiment. On the basis of single factor experiment, microwave power, extraction time and ethanol volume fraction were selected as the investigation factors. Five factors were set for each factor. The evaluation index of total flavonoids and total anthraquinone content was “normalized value” Optimum selection of microwave extraction process by using the method of star point design and response surface. Results: The optimum microwave extraction technology for the effective part of Morus alba to improve sleep was microwave power of 585 W, extraction time of 1 times, plus 10 times the amount of 82% ethanol extracted at 60 ℃ for 11 min. Conclusion: The optimized microwave-assisted extraction process is simple and predictable, providing experimental basis for follow-up formulation development and industrial production.
