观察苏木乙酸乙酯提取物对MsPGN模型大鼠肾组织TNF-α mRNA表达,探讨苏木免疫抑制作用机制。将40只大鼠随机分成四组,每组10只,复制邹氏[1]改良的慢性血清病MsPGN动物模型,空白组和模型组每日灌服按体重计算等容量橄榄油;雷公藤组给予雷公藤多甙片橄榄油灌胃,6.5 mg/kg.d;苏木组给予苏木橄榄油灌胃,1.15 g/kg.d(含生药量);观察相关指标。结果显示各组电泳条带亮度有明显差异,模型组与空白组比较(P<0.01);与模型组相比较,苏木组、雷公藤组肾脏中的TNF-α mRNA表达减少,苏木组(P<0.01),雷公藤组(P<0.05);而苏木组与雷公藤组比较无显著性差别(P>0.05)。综上可见苏木乙酸乙酯能够抑制大鼠MsPGN模型肾脏中的TNF-α mRNA表达,减轻其病理损伤,并有利尿消肿和降低蛋白尿的作用。
To observe the expression of TNF-α mRNA in renal tissue of MsPGN rat model with ethyl acetate extract of hematoxylin. Forty rats were randomly divided into four groups of 10 rats, and Chow’s [1] modified chronic serum disease MsPGN animal model was replicated. The blank group and the model group were fed daily by volume with equal volume of olive oil; triptolide group The rats were given intragastric administration of Tripterygium wilfordii olive oil at a dose of 6.5 mg/kg.d. The Sumu group was given intragastric administration of olive oil, 1.15 g/kg.d (including crude drug). Observed the relevant indicators. The results showed that there were significant differences in the brightness of the electrophoresis bands between the groups, and the model group was compared with the blank group (P<0.01). Compared with the model group, the expression of TNF-α mRNA in the kidney of the Sumu group and Tripterygium wilfordii group decreased, and the Sumu group (P<0.01), tripterygium wilfordii group (P<0.05); but there was no significant difference between the Sumitsu group and Tripterygium wilfordii group (P>0.05). In summary, it can be seen that ethyl palmitate can inhibit the expression of TNF-α mRNA in the kidney of rat MsPGN model, reduce its pathological damage, and have a beneficial effect on reducing urine swelling and proteinuria.
