目的:探讨大鼠胚胎胰腺发育外分泌功能基因表达调控. 方法:采用高密度寡核苷酸芯片(Affemetrix芯片)对E15.5和E18.5胚胎胰腺进行基因转录水平分析,基于获得的基因表达信息对UCSC,TRANSFAC,NCBI等公共数据库进行检索. 结果:在差异或特异表达的1319个基因中与胰腺细胞分化相关的转录因子和信号分子表达均下调,但胰腺外分泌部特有的转录因子PTF1-P48,Mistl表达显著上调,多种消化性酶表达亦显著增强,然而与分泌功能相关的VAMP-2却无表达. 结论:从E15.5至E18.5大鼠胚胎胰腺外分泌部积极地完善其消化功能,但尚不能分泌多种消化性酶.
OBJECTIVE: To investigate the regulation of exocrine function gene expression in rat embryonic pancreas.Methods: The gene transcription level of E15.5 and E18.5 embryos was analyzed by high-density oligonucleotide microarray (Affemetrix chip). Based on the obtained gene expression information The common databases of UCSC, TRANSFAC and NCBI were searched.Results: The expression of transcription factors and signaling molecules related to the differentiation of pancreatic cells were all down-regulated in 1319 differentially or specifically expressed genes, but PTF1-P48 , Mistl expression was significantly up-regulated, and the expression of various digestive enzymes was also significantly increased, while the secretion-related VAMP-2 was not expressed.Conclusion: The exocrine pancreas of E15.5 to E18.5 rats actively improved their digestion Function, but can not secrete a variety of digestive enzymes.
