目的探讨添加短链脂肪酸(SCFAs)的全肠外营养(TPN)对术后化疗大鼠结肠粘膜细胞增殖作用的影响。方法30只SD大鼠在无渣饮食2d后建立TPN和结肠吻合模型。术后随机分为TPN对照组(对照组)、化疗+TPN组(化疗组)、化疗+TPN+SCFAs组(SCFAs组),每组10只。进行实验5 d,术后第6 d留取标本采用流式细胞术分析结肠粘膜细胞周期,观察其增殖情况。结果化疗组结肠粘膜细胞增殖期百分比(6.23±2.75)%显著低于对照组[(9.81±2.78)%,P=0.014]和SCFAs组[(11.57±3.54)%,P=0.001];化疗组结肠粘膜细胞的增殖指数(11.87±2.61)%显著低于其他2组[(20.25±7.22)%,P=0.002;(19.70±5.12)%,P=0.003]。结论添加SCFAs的TPN能促进术后化疗大鼠结肠粘膜细胞的增殖。
Objective To investigate the effect of total parenteral nutrition (TPN) with short chain fatty acids (SCFAs) on the proliferation of colonic mucosal cells in rats after chemotherapy. Methods Thirty SD rats were established TPN and colon anastomosis models after 2 days without residue diet. The patients were randomly divided into TPN control group (control group), chemotherapy + TPN group (chemotherapy group) and chemotherapy + TPN + SCFAs group (SCFAs group). Five days after operation, samples were collected on the 6th day after operation, and the colonic mucosal cell cycle was analyzed by flow cytometry to observe the proliferation. Results The percentage of colonic mucosal cell proliferation in the chemotherapy group was significantly lower than that in the control group (6.23 ± 2.75% vs 9.81 ± 2.78%, P = 0.014 and SCFAs [11.57 ± 3.54]%, P 0.001] The proliferation index of colonic mucosal cells (11.87 ± 2.61)% was significantly lower than that of the other two groups [(20.25 ± 7.22)%, P = 0.002; (19.70 ± 5.12)%, P = 0.003]. Conclusion The addition of SCFAs TPN can promote the proliferation of colonic mucosal cells in rats after chemotherapy.
