目的:研究复合螺旋藻多糖对体外培养的人乳腺癌MB-231肿瘤细胞的抑制作用及可能的作用机制。方法:以人乳腺癌MB-231细胞株为研究对象,将螺旋藻多糖(PSP)与银杏叶提取物(GBE)按不同比例复合,并用不同浓度的复合制剂分别对细胞作用后,分别在48h、72h后,显微镜下观察细胞形态学变化,MTT法测其生长抑制率。48h后,PI及AnnexinV-FITC/PI染色,用流式细胞仪测其细胞周期及凋亡率。结果:复合螺旋藻多糖对人乳腺癌MB-231细胞均有不同程度的抑制作用,复合组优于单一成分组,抑制生长效果与时间和剂量呈依赖关系;将人乳腺癌MB-231细胞阻滞于G0/G1期,抑制肿瘤细胞的有丝分裂;促进肿瘤细胞凋亡,与空白组相比差异显著。结论:复合螺旋藻多糖通过阻断细胞周期达到抑制体外培养的人乳腺癌MB-231细胞的生长的目的,具有较好的抗肿瘤活性。
Objective: To study the inhibitory effect of polysaccharide from Spirulina platensis on human breast cancer cell line MB-231 and its possible mechanism. Methods: Human breast cancer cell line MB-231 was used as the research object. The polysaccharide of Spirulina platensis (GBE) and ginkgo biloba extract (GBE) were mixed in different proportions and treated with different concentrations of compound preparations respectively at 48h After 72 hours, the morphological changes of the cells were observed under a microscope. The growth inhibition rate was measured by MTT assay. After 48h, PI and Annexin V-FITC / PI staining were used to measure the cell cycle and apoptosis rate by flow cytometry. Results: The polysaccharide of Spirulina platensis could inhibit the growth of human breast cancer cell line MB-231 to some extent. The combination group was better than the single component group, and the growth inhibition effect was dependent on time and dosage. Stagnation in G0 / G1 phase, inhibition of tumor cell mitosis; promote tumor cell apoptosis, compared with the blank group significant difference. Conclusion: The polysaccharide of Spirulina platensis has good anti-tumor activity by blocking the cell cycle to inhibit the growth of human breast cancer MB-231 cells cultured in vitro.
