目的探讨汉滩病毒A9株(HTNVA9)对肾实质细胞的致病作用。方法采用免疫标记技术和电镜技术观察HTNVA9株对人胚肾小球系膜细胞(MsC)的感染情况和病毒唑、磷甲酸钠对病毒感染的抑制作用。结果HTNVA9株能感染MsC并能在其中增殖,形成包涵体;病毒唑和磷甲酸钠能抑制HTNVA9株对MsC的感染,且加药时间愈早,抑制作用愈强。结论汉滩病毒感染是引起肾综合征出血热肾实质直接损伤的重要机制之一;为临床早期抗病毒药物治疗提供了实验依据。
Objective To investigate the pathogenic effect of Hantaan virus A9 strain (HTNVA9) on renal parenchymal cells. Methods Immunofluorescence staining and electron microscopy were used to observe the infection of HTNVA9 strain on human mesangial cells (MsC) and the inhibitory effect of ribavirin and sodium phosphate on viral infection. Results The strain of HTNVA9 could infect MsC and proliferate into it, forming inclusion bodies. Ribavirin and sodium phosphate could inhibit the infection of MsC by HTNVA9 strains, and the sooner the dosing time was, the stronger the inhibition was. Conclusion Hantaan virus infection is one of the most important mechanisms that cause direct injury of renal pheochromocytoma in patients with hemorrhagic fever with renal syndrome. It provides an experimental basis for the early clinical treatment of antiviral drugs.
