目的:研究前体饲喂对高山红景天细胞生长和红景天苷含量的影响。方法:在细胞培养起始期分别添加苯丙氨酸、肉桂酸和酪氨酸3种前体,在细胞生长的稳定期第14 d收获细胞,测定细胞干重、红景天苷含量、多酚氧化酶(PPO)和苯丙氨酸裂解酶(PAL)的活性。结果:苯丙氨酸的最佳添加质量浓度为20 mg·L-1,肉桂酸和酪氨酸的最佳添加质量浓度均为10 mg·L-1,此时红景天苷含量分别为2.65,3.50,3.21 mg·g-1。但高浓度的酪氨酸对细胞的生长有一定的抑制作用。结论:3种前体在合适的浓度下均能促进细胞中红景天苷的生物合成,并提高多酚氧化酶(PPO)、苯丙氨酸裂解酶(PAL)的活性,其中肉桂酸作用明显。
Objective: To study the effect of precursor feeding on the growth of Rhodiola rosea and the content of salidroside. Methods: Three precursors of phenylalanine, cinnamic acid and tyrosine were added in the initial stage of cell culture, and the cells were harvested on the 14th day after the cell growth was stable. The cell dry weight, salidroside content, polyphenols Oxidase (PPO) and phenylalanine lyase (PAL) activity. Results: The optimal concentration of phenylalanine was 20 mg · L-1, and the optimal concentrations of cinnamic acid and tyrosine were 10 mg · L-1. The contents of salidroside were 2.65,3.50,3.21 mg · g-1. However, high concentrations of tyrosine cells have some inhibitory effect. CONCLUSION: All three precursors can promote the biosynthesis of salidroside in cells and increase the activity of polyphenol oxidase (PPO) and phenylalanine lyase (PAL), and the activity of cinnamic acid obvious.
