目的检测膀胱癌患者着色性干皮病F蛋白(XPF)的表达水平,并探讨其临床意义。方法Trizol法提取25份膀胱癌组织及10份癌旁正常组织总RNA,经RT-PCR扩增后,以扩增产物为模板,经实时荧光定量PCR法检测XPF的表达水平。结果各基因扩增效率近似相等,且具有很好的特异性,膀胱癌组织中XPF基因的表达水平明显低于癌旁正常组织。结论XPF可能参与了膀胱癌的发生和发展过程,对膀胱癌的转移潜能及预后具有一定意义。
Objective To detect the expression of F protein (XPF) in patients with chromosomal dry skin disease and to investigate its clinical significance. Methods Trizol method was used to extract total RNA from 25 samples of bladder cancer tissues and 10 normal tissues. After amplification by RT-PCR, the expression of XPF was detected by real-time fluorescence quantitative PCR. Results The amplification efficiency of each gene was approximately equal and had good specificity. The expression level of XPF gene in bladder cancer tissues was significantly lower than that in adjacent normal tissues. Conclusions XPF may be involved in the occurrence and development of bladder cancer, which may be of significance for the metastatic potential and prognosis of bladder cancer.
