目的:研究健脾活血方对胃癌前病变(PLGC)大鼠胃黏膜组织中环氧合酶-2(COX-2)及基质金属蛋白酶-2(MMP-2)表达的影响。方法:60只大鼠随机分为2组:正常组10只和造模组50只。正常组正常喂养,余下大鼠每天给予150 mg·L-1的甲基硝基亚硝基胍(MNNG)溶液自由饮用,0.03%雷尼替丁加入170 mg·L-1的MNNG溶液中按10 mL·kg-1每天灌胃1次,40%的乙醇按10 mL·kg-1每周灌胃2次,连续12周,其中第5周开始结合饥饱失常,建立胃癌前病变大鼠模型。将造模成功的40只大鼠随机分为模型组(0.9%氯化钠溶液)、胃复春组(0.86 g·kg-1)、健脾活血方高、中、低剂量组(32,16,8 g·kg-1),每组8只,每组每天给予不同药物灌胃一次,连续10周。实验第22周末处死大鼠,给予相应处理后,快速免疫组化检测COX-2及MMP-2表达情况。结果:与正常组相比,模型组COX-2,MMP-2表达均显著升高(P<0.01);与模型组相比,健脾活血方高、中、低剂量组、胃复春组COX-2,MMP-2表达均显著降低(P<0.01或P<0.05);与胃复春组相比,健脾活血方高剂量组COX-2,MMP-2表达均显著降低(P<0.05),中、低剂量组COX-2,MMP-2表达与胃复春组相比无明显差异。结论:健脾活血方治疗胃癌前病变与降低胃黏膜中COX-2及MMP-2的表达相关。
Objective: To investigate the effects of Jianpi Huoxue Recipe on the expression of cyclooxygenase-2 (COX-2) and matrix metalloproteinase-2 (MMP-2) in gastric mucosa of gastric precancerous lesions (PLGC) rats. Methods: Sixty rats were randomly divided into two groups: normal group (n = 10) and model group (n = 50). The rats in the normal group were fed with normal diet. The remaining rats were given 150 mg · L -1 MNNG solution daily for free drinking. When 0.03% ranitidine was added to 170 mg · L -1 MNNG solution, 10 mL · kg-1 orally daily, 40% ethanol 10 mL · kg-1 orally twice a week for 12 weeks, the first 5 weeks of the combination of starvation and disturbance, the establishment of gastric precancerous lesions in rats model. The 40 successful rats were randomly divided into model group (0.9% sodium chloride solution), Wei Fuchun group (0.86 g · kg-1), Jianpihuoxue Fang high, medium and low dose groups (32, 16,8 g · kg-1), each group of 8, each group given different drugs gavage once a week for 10 weeks. At the end of the experiment, the rats were sacrificed at the end of the 22nd week. After corresponding treatment, the expression of COX-2 and MMP-2 were detected by rapid immunohistochemistry. Results: Compared with the normal group, the expressions of COX-2 and MMP-2 in the model group were significantly increased (P <0.01). Compared with the model group, the expressions of COX-2, MMP- The expression of COX-2 and MMP-2 in high-dose Jianpi Huoxue prescription group was significantly lower than that in Fufuchun group (P <0.01 or P <0.05) 0.05). There was no significant difference in the expression of COX-2 and MMP-2 between moderate and low dose groups and Wei Fu Chun group. Conclusion: Jianpi Huoxue Fang treatment of gastric precancerous lesions and reduce the expression of COX-2 and MMP-2 in gastric mucosa.
