目的通过研究痛风患者载脂蛋白C-Ⅲ(ApoC-Ⅲ)启动子的单核苷酸多态性与高甘油三酯(HTG)血症之间的关系,探讨该基因变异与痛风病程进展的关系。方法测定中国山东地区300例痛风患者及300例健康人群中的ApoC-Ⅲ启动子的两个位点C-482T和T-455C的基因型和等位基因频率,同时测定两组人群的血脂水平,并对其进行比较。结果痛风组较对照组TG水平增高,差异有统计学意义(t=6.75,P<0.01)。455位点的CC、CT和TT基因型在痛风组和对照组中分布频率分别为23.0%、24.0%、53.0%和30.3%、13.3%、56.3%,差异有统计学意义(χ2=12.47,P<0.01);455位点等位基因C的分布频率在痛风组和对照组中49.5%和58.5%,差异有显著性统计学意义(χ2=9.78,P<0.01);482位点的CC、CT和TT基因型在痛风组和对照组中分布频率分别为28.0%、23.0%、49.0%和16.0%、33.7%、50.3%,差异有显著性统计学意义(χ2=15.89,P<0.01);482位点等位基因T的分布频率在痛风组和对照组中47.5%和58.8%,差异有统计学意义(χ2=15.47,P<0.01)。结论 ApoC-Ⅲ启动子的455和482位点的基因多态性与痛风患者的HTG发病有关。
Objective To investigate the relationship between single nucleotide polymorphisms of apolipoprotein C-Ⅲ (ApoC-Ⅲ) promoter and high triglyceride (HTG) hyperlipidemia in gout patients, relationship. Methods The genotype and allele frequencies of C-482T and T-455C at two sites of ApoC-Ⅲ promoter were determined in 300 gout patients and 300 healthy persons in Shandong Province, China. The levels of plasma lipids , And compare them. Results Compared with the control group, the gout group had higher TG levels, the difference was statistically significant (t = 6.75, P <0.01). The frequencies of CC, CT and TT genotypes at 455 loci were 23.0%, 24.0%, 53.0% and 30.3%, 13.3% and 56.3% in gout group and control group, respectively (χ2 = 12.47, P <0.01). The distribution frequency of allele C at position 455 was 49.5% and 58.5% respectively in gout group and control group (χ2 = 9.78, P <0.01) . The distribution frequencies of CT and TT genotypes in gout group and control group were 28.0%, 23.0%, 49.0% and 16.0%, 33.7% and 50.3%, respectively, with significant difference (χ2 = 15.89, P <0.01) ). The distribution frequency of allele T at 482 locus was 47.5% and 58.8% respectively in gout group and control group (χ2 = 15.47, P <0.01). Conclusion The gene polymorphisms at 455 and 482 of ApoC-Ⅲ promoter are associated with the pathogenesis of HTG in gout patients.
