目的:探讨桑叶多糖(mulberry leaf polysaccharide,MLP)对糖尿病肾病(Diabetic nephropathy,DN)大鼠的保护作用。方法:经筛选后将Wistar大鼠随机分为正常对照组、模型组、MLP低、中、高剂量治疗组。采用一次性腹腔注射链脲佐菌素(STZ)的方法建立DN大鼠模型。给予不同剂量MLP治疗8W后,分别检测大鼠空腹血糖(FPG)、尿液中微量白蛋白(m Alb)含量、血清中尿素氮(BUN)、肌酐(Cr)含量;应用免疫组化法检测肾脏转化生长因子TGF-β1蛋白的表达、RT-PCR法检测IRS-1 mRNA的表达。结果:MLP能显著降低DN大鼠FPG、m Alb、BUN、Cr含量(P<0.01);MLP能明显上调DN大鼠肝组织IRS-1 mRNA表达(P<0.01),亦能下调DN大鼠肾脏TGF-β1蛋白的表达(P<0.01)。结论:MLP对DN大鼠的肾脏具有一定肾保护作用,其作用机制可能是通过上调DN大鼠肝组织IRS-1 mRNA表达及下调TGF-β1蛋白表达来实现。
Objective: To investigate the protective effect of mulberry leaf polysaccharide (MLP) on diabetic nephropathy (DN) rats. Methods: After screening, Wistar rats were randomly divided into normal control group, model group and MLP low, medium and high dose treatment groups. A rat model of DN was established by intraperitoneal injection of streptozotocin (STZ). After being treated with different doses of MLP for 8 weeks, fasting plasma glucose (FPG), urine microalbumin (M Alb) and serum BUN and Cr were measured respectively. Immunohistochemical The expression of transforming growth factor-β1 (TGF-β1) in kidney and the expression of IRS-1 mRNA were detected by RT-PCR. Results: MLP significantly decreased the levels of FPG, m Alb, BUN and Cr in DN rats (P <0.01); MLP significantly increased IRS-1 mRNA expression in liver of DN rats (P <0.01) Kidney TGF-β1 protein expression (P <0.01). CONCLUSION: MLP has a protective effect on the kidneys of DN rats. Its mechanism may be through up-regulating IRS-1 mRNA expression and down-regulating the expression of TGF-β1 in the liver of DN rats.
