目的探讨滋补脾阴方药对脊髓继发性损伤的保护作用。方法采用Allen法致伤造成家兔脊髓损伤模型,随机分为假手术组,2h、12h、24h、60h滋补脾阴方药组及各中药组生理盐水对照组,观察脊髓细胞线粒体Na+,K+-ATP酶,Ca2+,Mg2+-ATP酶活性及超氧化物歧化酶(SOD)和丙二醛(MDA)含量变化。结果脊髓损伤后脊髓细胞线粒体Na+,K+-ATP酶、Ca2+,Mg2+-ATP酶活性、SOD含量明显降低,MDA含量明显升高。使用滋补脾阴方药后,增高Na+,K+-ATP酶、Ca2+,Mg2+-ATP酶活性,使SOD含量升高,使MDA含量降低。结论滋补脾阴方药可以改善脊髓细胞线粒体ATP酶活性,缓解脂质过氧化反应,从而发挥脊髓保护作用。
Objective To investigate the protective effect of nourishing spleen-yin decoction on secondary injury of spinal cord. Methods Spinal cord injury model induced by Allen method was randomly divided into sham operation group, nourishing spleen-yin decoction group and normal saline control group at 2h, 12h, 24h, 60h. Spinal cord mitochondria Na+, K+-ATP were observed. Activities of enzymes, Ca2+, Mg2+-ATPase, superoxide dismutase (SOD) and malondialdehyde (MDA) content. Results After spinal cord injury, the activity of Na+, K+-ATPase, Ca2+, Mg2+-ATPase and SOD in mitochondria of spinal cord cells were significantly decreased, and the content of MDA was significantly increased. After use of nourishing spleen and yin herbs, Na+, K+-ATPase, Ca2+, Mg2+-ATPase activity increased, SOD content increased, and MDA content decreased. Conclusion Nourishing spleen and Yin herbs can improve mitochondrial ATPase activity of spinal cord cells, relieve lipid peroxidation, and thus play a role in spinal cord protection.
