【目的】建立宽体金线蛭油酸的气相色谱指纹图谱。【方法】以不同来源地和不同规格的共10批宽体金线蛭为研究对象,选择Agilent 6890N型气相色谱仪,DB-WAX色谱柱(30 mm×0.32 mm×0.25μm),气化温度为270℃,柱温130℃,检测器(FID)温度为280℃。采用中国药典委员会颁布的《中药色谱指纹图谱相似度评价系统2004A》进行相似度计算。【结果】不同规格的宽体金线蛭油酸含量差异有统计学意义(F2,7=7.350,P=0.019),清水晒干品分别和矾品、饮片/晒干的油酸含量比较差异有统计学意义(P=0.021,P=0.009)。指纹图谱的相似度在0.458~0.998之间,其中与其他样品和对照指纹图谱的相似度比较,来源于广西荔浦的样品相似度为最低。【结论】大部分样品指纹图谱的相似度非常高,所建立的气相色谱指纹图谱可有效鉴别宽体金线蛭的优劣品,该方法可为宽体金线蛭的日常质量控制提供一定参考。
【Objective】 The aim of this study was to establish a gas chromatographic fingerprinting profile of Hirudin acid. 【Method】 A total of 10 batches of Hirudo verrucosus collected from different origins and different specifications were selected as the experimental materials. The chromatographic column of Agilent 6890N, DB-WAX (30 mm × 0.32 mm × 0.25 μm), gasification temperature 270 ° C, column temperature 130 ° C, detector (FID) temperature 280 ° C. The similarity was calculated by “Chinese Medicines Chromatographic Fingerprint Similarity Evaluation System 2004A” promulgated by the Chinese Pharmacopoeia Commission. 【Result】 The results showed that there were significant differences in the contents of oleic acid among different specifications of wide-body gold line (F2,7 = 7.350, P = 0.019) There was statistical significance (P = 0.021, P = 0.009). The similarity of fingerprints between 0.458 ~ 0.998, which compared with other samples and control fingerprints of similarity, samples from Lipu, Guangxi, the lowest similarity. 【Conclusion】 The fingerprint similarity of most samples is very high. The gas chromatographic fingerprinting established by this method can effectively identify the advantages and disadvantages of wide-bodied gold leeches. This method can provide reference for the daily quality control of barks .
