通过皮下注射四氯化碳复制化学性肝纤维化大鼠模型,分别于注射后4、8、12周处死动物。电镜下观察贮脂细胞的变化,免疫组化法检测Ⅳ型胶原在肝内分布的变化。结果表明,贮脂细胞在肝纤维化过程中经历了一个动态的变化,逐渐拉长、变形,成为肌成纤维样细胞,并分泌大量胶原。Ⅳ型胶原是肝纤维化中形成毛细血管化的主要胶原,它在肝内的沉积构成肝硬化门脉高压的病理学基础
The rats were sacrificed at 4, 8 and 12 weeks after injection by subcutaneous injection of carbon tetrachloride into a rat model of chemical liver fibrosis. The changes of the storage fat cells were observed under electron microscope. The distribution of type Ⅳ collagen in the liver was detected by immunohistochemistry. The results showed that the fat-storing cells undergone a dynamic change during liver fibrosis and gradually elongated and deformed to become myofibroblast-like cells and secrete a large amount of collagen. Collagen type IV is the major form of capillaries in liver fibrosis, and its deposition in the liver constitutes the pathological basis of cirrhosis and portal hypertension
