目的:探讨磁导向甲氨蝶呤缓释药物(FM-MTX)对舌癌细胞凋亡的诱导作用,了解新药FM-MTX治疗舌癌的作用机理。方法:选用不同剂量的FM-MTX与人舌鳞癌Tca8113细胞系共同培养。按不同时间收集细胞悬液,并在透射电镜下观察细胞超微结构的变化。结果:细胞经FM-MTX药物处理组,1h后,透射电镜下即可看到瘤细胞溶酶体大量增生,4h后瘤细胞胞浆溶酶体开始吞噬磁性微粒,24h吞噬颗粒明显增多,溶酶体膜开始裂解,颗粒释放到胞浆内。72h后其内质网及线粒体内也可见颗粒沉积,溶酶体膜有消失现象,呈现胞浆空泡结构。96h后部分瘤细胞出现凋亡。结论:甲氨蝶呤缓释药物(FM-MTX)对舌癌细胞凋亡起诱导作用,进入瘤细胞的细胞器,对肿瘤细胞有明显的杀伤作用
OBJECTIVE: To investigate the induction of apoptosis induced by magnetic-guided methotrexate sustained-release drug (FM-MTX) in tongue cancer cells and to understand the mechanism of action of FM-MTX in the treatment of tongue cancer. Methods: Different doses of FM-MTX were selected and co-cultured with Tca8113 human tongue squamous cell carcinoma cell line. The cell suspensions were collected at different times and the ultrastructural changes of the cells were observed under transmission electron microscope. Results: The cells were treated with FM-MTX. After 1h, the lysosomes of the tumor cells were observed under transmission electron microscope. After 4h, the cytosolic lysosomes began to phagocytize the magnetic particles, and the phagocytic granules were obviously increased after 24h The lysosome begins to lyse and the particles are released into the cytoplasm. 72h after the endoplasmic reticulum and mitochondria also showed deposition of particles, the lysosomal membrane disappears, showing cytoplasmic cavitation structure. Some tumor cells apoptosis after 96h. CONCLUSION: The methotrexate sustained-release drug (FM-MTX) can induce the apoptosis of tongue cancer cells and enter into the organelles of tumor cells with obvious killing effect on tumor cells
