目的 观察抗Hp多克隆羊奶抗体的体外被动免疫保护作用。 方法 用MTT法评价羊奶抗体中和Hp细菌本身及其超声裂解蛋白所致Hep - 2细胞空泡变性的细胞毒素作用效果。结果 抗Hp免疫乳抗体能够中和细菌悬液和超声裂解菌体蛋白诱导的Hep - 2细胞毒活性。免疫乳抗体 1∶8、1∶16稀释后能完全阻断细菌及超声裂解抗原对Hep - 2细胞的毒性作用 ,即与阴性对照组相比无显著性差异。经 1∶32~ 1∶2 5 6稀释能一定程度上阻断细胞毒作用 ,即与致细胞发生 5 0 %死亡的阳性对照相比有显著性差异 (P <0 0 5 )。结论 抗Hp免疫乳抗体在中和细菌毒素方面发挥一定的作用。提示局部应用抗体制剂作为被动免疫 ,有可能成为治疗Hp感染性疾病的有效方法之一。
Objective To observe the passive immunoprotection of anti-Hp polyclonal goat’s milk antibody in vitro. Methods MTT assay was used to evaluate the effect of goat milk antibody on the cytotoxicity of vacuolar degeneration of Hep - 2 cells induced by Hp bacteria and their sonicated lysates. Results The anti - Hp immune milk antibody could neutralize Hep - 2 cytotoxicity induced by bacterial suspension and sonication. Immune milk antibody 1: 8 and 1: 16 dilution completely blocked the toxic effects of bacteria and sonicated antigen on Hep - 2 cells, that is, no significant difference compared with the negative control group. Dilution 1:32 ~ 1:25 56 blocked the cytotoxicity to some extent, which was significantly different from that of the positive control group (P <0.05). Conclusion Anti-Hp immune milk antibodies play a role in the neutralization of bacterial toxins. Prompt topical application of antibody preparations as passive immunization, may become one of the effective ways to treat Hp infectious diseases.
