目的探讨促生长激素神经肽家族新成员Alarin对心血管系统生理功能是否具有调节作用,并为进一步认识Galanin家族分子的生物学作用提供更多的实验依据。方法选用3种不同来源的Alarin(1-25,3-25,6-25)分别来自Kofler实验室赠送的Alarin(1-25,3-25),国内上海强耀生物技术公司合成的Alarin(1-25,3-25)和第四军医大学基因所合成的Alarin(1-25,3-25,6-25),将实验大鼠随机分为2组:阳性对照组〔去甲肾上腺素(NE)组〕和实验组(Alarin组),通过整体动物实验测定血流动力学和离体心脏灌流实验测定血管张力,分别观察Alarin对实验大鼠心脏功能、血压、心率、血管张力等指标的影响。结果在实验组中3种不同来源的Alarin(1-25,3-25,6-25)分别在不同剂量时对大鼠的收缩压、舒张压、心率以及左室收缩压、左室收缩压上升和下降最大速率等心脏功能指标均没有明显影响,且不同终浓度的Alarin对大鼠腹主动脉的张力也无明显影响。而在NE组中,等剂量NE可明显增加收缩压、舒张压、心率以及左室收缩压、左室收缩压上升和下降最大速率等等心脏功能指标,且对大鼠腹主动脉具有明显的收缩作用。结论 Alarin对大鼠心脏及大血管的基础生理功能没有调节作用。
Objective To investigate whether Alarin, a new member of the growth hormone neuropeptide family, regulates cardiovascular system physiological functions and provide more experimental evidence for further understanding of the biological role of Galanin family molecules. Methods Three different sources of Alarin (1-25,3-25,6-25) were selected from Alarin presented by Kofler Laboratories (1-25,3-25), Alarin (produced by Shanghai Qianguang Biotechnology Co., Ltd.) 1-25,3-25) and Alarin synthesized by the Fourth Military Medical University (1-25,3-25,6-25). The experimental rats were randomly divided into two groups: positive control group (norepinephrine (Alarin group) and experimental group (Alarin group). In the NE group, the equal dose of NE can significantly increase the systolic blood pressure, diastolic blood pressure, heart rate and left ventricular systolic pressure, systolic blood pressure and maximum rate of decline and so on heart function index, and the rat abdominal aorta has obvious Shrinkage. Conclusion Alarin has no effect on the basic physiological functions of rat heart and macrovascular.
