为了探讨人超氧化物歧化酶 (SOD)值测定 ,作为一种新的细胞毒活性的检测方法及其临床应用的可行性 ,本文对含有不同细胞数的正常人外周血单个核细胞 ,反复冻融后 ,用放免方法 (RIA)测定上清中的SOD值 ,观察四组不同浓度细胞数量与所测SOD值的相关性。测定 2 0份标本SOD值均值±标准差分别为 :细胞数为 4× 10 6组 :SOD值为 4 90± 2 6 .3ng/mL ;2× 10 6组 ;2 4 9± 2 0 .8ng/mL ;1× 10 6组 :132 .5 5± 13.0 3ng/mL ;5× 10 5组 :10 2 .6 5±4 8.5ng/mL。四组细胞数与SOD均值的相关系数为 0 .91,绘制散点图 ,发现 5× 10 5组的均值偏离另三组的直线趋势 ,予以去除后 ,剩余的三组的相关系数为 1.0 3,呈现完全相关。结果显示 :SOD值在同一类型细胞中的含量相对恒定 ,在个体间一致性较高 ;SOD值与单个核细胞量存在直线相关性 ,提示这个参数可用于了解某类细胞被破坏的情况。值得注意的是 ,在细胞浓度较低时 ,SOD值可能会存在误差 ,建议实验时标本的细胞数宜控制在 1× 10 6以上
In order to investigate the determination of human superoxide dismutase (SOD), as a new cytotoxicity assay and the feasibility of its clinical application, we studied normal human peripheral blood mononuclear cells containing different numbers of cells, After thawing, the superoxide dismutase (SOD) in the supernatant was determined by radioimmunoassay (RIA), and the correlation between the number of cells in four different concentrations and the measured SOD value was observed. The mean ± standard deviation (SOD) of 20 samples were determined as follows: the number of cells was 4 × 10 6, the SOD value was 4 90 ± 2 6 .3ng / mL, 2 × 10 6 group, 2 4 9 ± 2 0.8g / mL; 1 × 10 6 groups: 132.55 ± 13.0 3 ng / mL; 5 × 10 5 groups: 10 2 .65 ± 4 8.5 ng / mL. The correlation coefficient between the four groups of cells and the average value of SOD was 0.91. The scatter plot was drawn and the average of 5 × 10 5 groups deviated from the straight-line trend of the other three groups. After the removal, the correlation coefficient of the remaining three groups was 1.0 3 , Is completely related. The results showed that the content of SOD was relatively constant in the same type of cells and the consistency was high among individuals. There was a linear correlation between the SOD value and the amount of mononuclear cells, suggesting that this parameter can be used to understand the destruction of certain types of cells. It is worth noting that SOD may be inaccurate at low cell concentration. It is suggested that the number of cells in the sample be controlled to be 1 × 10 6 or more
