目的探讨一种在染色体制备过程中避免样本间互相混淆污染、缩短制备时间和降低制备成本的简单方便、经济安全的方法。方法广西壮族自治区妇幼保健院从2005年起,对1000例血标本、1000例组织培养标本包括羊水、绒毛和流产组织分别使用传统法和一管法进行对照培养和制备染色体。结果两种方法成功率接近,均获得满意的染色体分裂相;但与传统法相比,运用一管法可以在染色体收获过程中未发生标本混淆的差错事故,并且平均每例血标本收获时间可以节约1.5 min,成本降低11.1元;每例组织标本可以节约12 min,成本降低74.4元。结论一管法也许是一种简便、安全、误差少和成本低值得推广的染色体制备方法。
Objective To explore a simple, economical and safe method to avoid confusion between samples, shorten the preparation time and reduce the preparation cost in the process of chromosome preparation. Methods From 2005 onwards, 1000 cases of blood samples, 1000 cases of tissue culture specimens including amniotic fluid, villus and abortion tissue were cultured and prepared by traditional method and one-tube method respectively in the MCHMC of Guangxi Zhuang Autonomous Region. Results The success rates of the two methods were close, and satisfactory chromosomal cleavage phase was obtained. However, compared with the traditional method, one-tube method could avoid mistakes in specimen confusion during chromosome harvesting, and the average harvest time of each blood sample could be saved 1.5 min, the cost reduced by 11.1 yuan; each tissue sample can save 12 min, the cost reduced by 74.4 yuan. Conclusion One tube method may be a simple, safe, less error and low cost to promote chromosome preparation methods.
