目的:建立高效液相色谱法测定凤仙透骨草中东莨菪素含量。方法:采用反相Alltima C18(4.6mm×250mm,5μm)色谱柱,流动相为甲醇-0.1%醋酸溶液(30:70),流速1.0mL.min-1,检测波长345nm,柱温35℃,进样量20μL。结果:东莨菪素线性范围为39.4~591ng(r=0.9997),检测限(S/N=3)为0.0748ng,平均回收率(n=9)为98.05%。结论:该方法操作简单、准确,灵敏度高,可作为凤仙透骨草中东莨菪素含量的分析方法。
OBJECTIVE: To establish a method for the determination of scopolamine in Perilla Fragrans by HPLC. METHODS: The mobile phase was methanol-0.1% acetic acid solution (30:70) at a flow rate of 1.0 mL · min-1 with a reversed phase Alltima C18 column (4.6 mm × 250 mm, 5 μm) Injection volume 20μL. Results: The linear range of scopolamine was 39.4-591 ng (r = 0.9997). The detection limit (S / N = 3) was 0.0748 ng. The average recovery was 98.05%. Conclusion: The method is simple, accurate and sensitive and can be used as an analytical method for the content of scopolamine in the Osmanthus fragrans.
