目的 :探讨脑缺血早期 (2 4h内 )大脑皮质星形胶质细胞 (Ast)的变化规律。方法 :应用胶质原纤维性酸性蛋白免疫组织化学ABC技术。结果 :缺血 1 5min和 3 0min组 ,缺血中心区大脑皮质胶质原纤维性酸性蛋白阳性细胞较均匀地分布于II~VI层 ,细胞数量明显多于非缺血区 ;缺血 1h和 2h组胶质原纤维性酸性蛋白阳性细胞数量进一步增加 ,胞质染色加深 ,并集中出现于III、IV层 ;缺血 3h组胶质原纤维性酸性蛋白阳性细胞进一步增大呈气球样 ,突起增长变粗 ,突起内出现水肿泡 ;缺血 6h组胶质原纤维性酸性蛋白细胞固缩 ,边界不清 ;1 2h和 2 4h组缺血中心区胶质原纤维性酸性蛋白细胞消失。同时观察到缺血 3h和 6h组缺血边缘区胶质原纤维性酸性蛋白细胞数量增多 ,直径增大 ,并可见到细胞分裂现象。结论 :星形胶质细胞对脑缺血早期神经元损伤具有较强的保护作用。
Objective: To investigate the changes of cerebral cortex astrocytes (Ast) in the early stage of cerebral ischemia (within 24 hours). Methods: The glial fibrillary acidic protein immunohistochemical ABC technique was used. Results: In the ischemic group at 15 min and 30 min, the number of glial fibrillary acidic protein positive cells in the cerebral cortex was more evenly distributed in the layer II-VI than in the non-ischemic area; The quantity of glial fibrillary acidic protein positive cells in 2h group was further increased, the cytoplasm staining deepened and appeared in III and IV layers. The glial fibrillary acidic protein positive cells in 3h group were further increased to balloon-like, The thickening of the protuberances and the appearance of water-and-foam bubbles in the protuberances; the glial fibrillary acidic protein cells in the ischemic 6h group shrank and the boundaries were unclear; the glial fibrillary acidic acidic protein cells in the ischemic center disappeared in 12h and 24h groups. At the same time, the number of glial fibrillary acidic protein cells in ischemic marginal zone was increased and the diameter increased at 3h and 6h after ischemia, and cell division was observed. Conclusion: Astrocytes have a strong protective effect on neurons in early stage of cerebral ischemia.
