目的:建立桦菌芝中氨基酸的含量测定方法。方法:采用阳离子交换色谱柱分离17种氨基酸,通过柱后衍生化检测分析。色谱柱为磺酸基强酸性阳离子交换树脂LCAK06/Na型(4.6 mm×150 mm),采用线性梯度洗脱,其中A泵(溶液泵)流速为0.45 mL.min-1,M泵(茚三酮泵)流速为0.25 mL.min-1;检测波长为440 nm(脯氨酸)和570 nm(其余氨基酸);进样量为50μL。结果:该方法中各氨基酸的线性关系良好;精密度RSD范围0.28%~1.67%,重复性RSD范围0.50%~2.13%;平均回收率为98.7%。结论:本研究建立的分析方法准确,灵敏度高,具有良好的重复性和回收率,可作为中药材中氨基酸的定量分析和研究方法。
Objective: To establish a method for determining the content of amino acids in gallbladder. Methods: Seventeen kinds of amino acids were separated by cation exchange chromatography and analyzed by column derivatization. The column was eluted with a linear gradient of LCAK06 / Na (4.6 mm × 150 mm) using a sulfonic acid-based strong cation exchange resin. The flow rate of A pump (solution pump) was 0.45 mL.min-1, Ketone pump) at a flow rate of 0.25 mL.min-1; detection wavelength was 440 nm (proline) and 570 nm (remaining amino acids); injection volume was 50 μL. Results: The linearity of each amino acid in this method was good. The precision of RSD ranged from 0.28% to 1.67%, and the range of RSD was 0.50% to 2.13%. The average recovery was 98.7%. Conclusion: The analytical method established in this study is accurate, sensitive and has good repeatability and recovery rate. It can be used as a quantitative analysis and research method of amino acids in traditional Chinese medicine.
