目的 比较在大鼠脑发育过程中 ,Wnt 1在喙侧神经干细胞迁移流中表达水平的时空差异。方法 以 5 溴脱氧尿嘧啶尿苷 (BrdU)对胚胎 14d、出生后 0d和 7d的Wistar大鼠行活体标记 ,取各时相点鼠脑 ,冰冻切片 ,行BrdU、Wnt 1和胶质纤维酸性蛋白 (GFAP)免疫荧光染色 ,比较在上述三个时相点 ,Wnt 1于室管膜前下区、迁移流主干部分和嗅球三个部位的表达情况。结果 在时间上 ,Wnt 1的表达在胚胎 14d高于出生后 0d和 7d ,出生后 0d和 7d的表达情况相似 ;在空间上 ,Wnt 1的表达在嗅球最高 ,室管膜前下区稍低 ,在神经干细胞迁移流主干部分的表达最低。结论 随着大鼠脑的发育 ,Wnt 1于喙侧神经干细胞迁移流中的表达逐渐减弱 ;在胚胎期及出生后早期 ,Wnt 1在喙侧神经干细胞迁移流中表达的空间分布情况相似。
Objective To compare the spatial and temporal differences in the expression of Wnt 1 in the migratory flow of the rostral neural stem cells during rat brain development. Methods Wistar rats were stained with bromodeoxyuridine (BrdU) for 14 days and 0 and 7 days after birth. The brains of rats were sacrificed and frozen sections were harvested at each time point. BrdU, Wnt 1 and glial fibrillary acidic (GFAP) immunofluorescence staining. The expression of Wnt 1 in the anterior ependymal zone, the migratory part of the flow and the olfactory bulb at the three time points were compared. Results The expression of Wnt 1 in the embryo at 14 d was higher than that at 0 d and 7 d after birth, and the expression of Wnt 1 was similar at 0 d and 7 d after birth. The spatial distribution of Wnt 1 was highest in the olfactory bulb and slightly lower in the anterior ependymal zone , With the lowest expression in the stem of neural stem cell migration stream. Conclusions With the development of rat brain, the expression of Wnt 1 in the migrating stream of beaked side neural stem cells gradually weakened. The distribution of Wnt 1 in the migration flow of beaked side neural stem cells was similar between embryonic and early postnatal stages.
