目的以海蛇全体干品为原料,海蛇水解蛋白得率为指标,优选酶法水解海蛇蛋白的工艺。方法对比研究了温度、酶量、时间、固液比对木瓜蛋白酶、枯草杆菌中性蛋白酶、胰蛋白酶、碱性蛋白酶酶解海蛇蛋白的影响,以正交试验优化酶解条件。结果枯草杆菌中性蛋白酶水解海蛇蛋白的得率约为42%,其最佳条件为温度50℃,pH值7.0,固液比1∶9(g/ml),酶量3000 U/3g(原料),时间7 h。木瓜蛋白酶、胰蛋白酶、碱性蛋白酶酶解得率分别为36.2%、38.8%、38.8%。结论枯草杆菌中性蛋白酶水解海蛇蛋白的得率最高。
OBJECTIVE To obtain the enzymatic hydrolysis of sea snake protein by using the whole dried sea snake as raw material and the yield of sea snake hydrolyzed protein as an index. Methods The effects of temperature, amount of enzyme, time, ratio of solid to liquid on papain, B. subtilis neutrase, trypsin and alkaline protease on the hydrolysis of sea snake protein were studied. The conditions of enzymolysis were optimized by orthogonal test. Results Bacillus subtilis neutral protease hydrolyzed sea snake protein yield was about 42%. The optimal conditions were temperature 50 ℃, pH 7.0, solid-liquid ratio 1: 9 (g / ml) and enzyme amount 3000 U / ), Time 7 h. The yields of papain, trypsin and alkaline protease were 36.2%, 38.8% and 38.8%, respectively. Conclusion Bacillus subtilis neutral protease has the highest yield of sea snake protein.
