目的:观察外源性超氧化物歧化酶(superoxidedismutase,SOD)和过氧化氢酶(catalase,CAT)对培养的人视网膜色素上皮(retinalpigmentepithelium,RPE)细胞光化学损伤的保护作用。方法:将铺满期原代培养的人RPE细胞分为三组。A组直接作光照处理,B组在培养液中加入SOD(90U/ml)和CAT(900U/ml)后30分钟进行光照,C组为对照组。光照强度2400Lx,时间6小时。处理结束后将三组细胞以2.4×105/ml的密度分别接种于24孔板,每孔0.4ml。于接种后1、3、5和7天观察细胞形态,并进行细胞计数。结果:光照后3天时,A、B组细胞生长缓慢,C组细胞生长活跃;3天后B组细胞生长接近C组,比A组明显活跃(P<0.05)。结论:可见光可使培养的人RPE细胞生长能力降低,外源性SOD和CAT对细胞的损伤具有保护作用。
Objective: To observe the protective effects of exogenous superoxidedismutase (SOD) and catalase (CAT) on the photochemical damage of cultured human retinal pigment epithelium (RPE) cells. Methods: The primary cultured human RPE cells were divided into three groups. Group A was exposed to light immediately. Group B was exposed to light (90 U / ml) and CAT (900 U / ml) for 30 minutes in group B, and control group. Light intensity 2400Lx, time 6 hours. After the treatment, three groups of cells were inoculated into 24-well plates at a density of 2.4 × 10 5 / ml, respectively, at a volume of 0.4 ml per well. Cell morphology was observed 1, 3, 5 and 7 days after inoculation and cell counts were performed. Results: After 3 days, the cells in group A and B grew slowly and the cells in group C grew actively. After 3 days, the cell growth in group B was close to that in group C, which was significantly more active than that in group A (P <0.05). CONCLUSION: Visible light can reduce the growth ability of cultured human RPE cells, and exogenous SOD and CAT have a protective effect on cell injury.
