目的:建立一种高灵敏度的液质联用(C-MS/MS)快速测定方法,用于快速测定大鼠血浆中回拉西坦的浓度,并计算了回拉西坦在大鼠体内的药代动力学参数。方法:血浆样品加乙腈沉淀蛋白,以乙腈-水(68:32)为流动相,用Apollo C_(18)色谱柱(4.6mm×150mm,5m)分离,采用电喷雾离子化源(ESI)三重四级杆串联质谱,以选择离子监测(SIM)方式进行检测。结果:回拉西坦线性范围为:0.0222~2.22gmL~(-1),回归方程为:A=301733+2×10~(7)C,R~2=0.9949(n=7);低、中、高3个质量浓度的平均回收率分别为:99.55%、99.10%、98.65%。结论:该法快速、灵敏、专属,适用于测定生物样本中回拉西坦的血药浓度。
OBJECTIVE: To establish a high-sensitivity liquid chromatography-mass spectrometry (LC-MS / MS) rapid determination method for the rapid determination of the concentration of carbenicillin in rat plasma and to calculate the concentration of carbachol in rats Pharmacokinetic parameters. Methods: The plasma samples were precipitated with acetonitrile and separated on an Apollo C 18 column (4.6 mm × 150 mm, 5 m) using acetonitrile-water (68:32) as the mobile phase. The electrospray ionization source (ESI) triple Quadrupole rod tandem mass spectrometry was performed using a selective ion monitoring (SIM) method. Results: The linear range of cecilantin was 0.0222 ~ 2.22gmL ~ (-1), the regression equation was: A = 301733 + 2 × 10 ~ (7) C, R ~ 2 = 0.9949 (n = 7) The average recoveries of the three medium and high concentrations were 99.55%, 99.10% and 98.65%, respectively. Conclusion: The method is rapid, sensitive and specific and is suitable for the determination of the plasma concentration of rehracetam in biological samples.
