目的:研究3种不同萃取方式对太子参萃取物的抗炎症反应功效的差异,为太子参在抗炎症方面的应用提供参考。方法:分别采用水加热回流萃取,95%乙醇超声波萃取和超临界萃取法萃取太子参获得不同萃取物,利用MTT实验评估萃取物的安全性,脂氧合酶活性抑制试验和一氧化氮清除实验评估太子参萃取物的抗炎活性,运用p UC119 DNA质粒模型评估太子参萃取物的DNA保护作用。结果:太子参水萃取物和醇萃取物具有显著抑制脂氧合酶活性,活性由强至弱的顺序为水萃取物、醇萃取物;3种方式所得太子参萃取物均具有清除NO活性,活性由强至弱的顺序为水萃取物、超临界萃取物、醇萃取物;太子参水萃取物和醇萃取物均具有一定的DNA保护作用。结论:太子参萃取物抗应激损伤与其具有抑制脂氧合酶活性,清除NO和DNA保护功能有关。
OBJECTIVE: To study the difference in the anti-inflammatory effects of three different extraction methods on the extract of Radix Pseudostellariae, and provide a reference for the application of Radix Pseudostellaria in anti-inflammatory. Methods: Extracts were extracted by water heating reflux extraction, ultrasonic extraction by 95% ethanol and supercritical extraction, respectively. The safety of extracts, inhibition of lipoxygenase activity and nitric oxide depletion were evaluated by MTT assay Assess the anti-inflammatory activity of the extract of Pseudostellaria heterophylla, and evaluate the DNA protection effect of the extract of Pseudostellaria polymorpha using the pUC119 DNA plasmid model. Results: The water extract and alcohol extract of Pseudostellaria heterophylla had a significant inhibitory effect on the activity of lipoxygenase, and the order of activity from strong to weak was water extract and alcohol extract. The extracts of Pseudostellaria heterophylla had NO scavenging activity, The order of activity from strong to weak was water extract, supercritical extract and alcohol extract. The water extract and alcohol extract of P. heterophylla had certain DNA protection. Conclusion: The anti-stress injury of Taizishen extract is related to the inhibition of lipoxygenase activity and NO and DNA protection.
