高脂血清取自实验性高脂血症的家兔。平滑肌细胞(SMC)取自家兔主动脉,并用含20%小牛血清的培养基培养。用流式细胞术(FCM)对SMC增生各期的DNA含量进行测定,研究5%高脂血清在24、36和48小时,0.5mg/L异搏定在48小时对SMC周期进程的影响。结果表明,高脂血清可使更多的SMC从G_0/G_1期进入S期及G_2/M期,从而促进SMC增生;异搏定则抑制上述过程。
High-fat serum was obtained from rabbits with experimental hyperlipidemia. Smooth muscle cells (SMC) were taken from rabbit aorta and cultured in medium containing 20% calf serum. Flow cytometry (FCM) was used to determine the DNA content of SMC hyperplasia in each phase. The effects of 5% hyperlipidemia serum at 24, 36, and 48 hours and 0.5 mg/L verapamil on SMC cycle progression at 48 hours were studied. The results showed that high-lipid serum increased the number of SMCs from G 0 /G 1 phase to S phase and G 2 /M phase, which promoted SMC hyperplasia; and verapamil inhibited the above process.
