目的考察灌胃给药华卟啉钠在大鼠体内的主要代谢产物和代谢途径。方法收集华卟啉钠灌胃后大鼠的血浆、尿液和粪便,采用Thermo Q Exactive Orbitrap四极杆-轨道阱高分辨质谱仪对大鼠尿、粪及血浆中的代谢产物进行研究,通过获得的精确分子量、二级碎片图谱,推断SD大鼠尿、粪及血浆中的代谢物,并绘制华卟啉钠在SD大鼠体内的代谢途径。结果在大鼠的尿、粪及血浆样品中共检测到10个代谢产物,主要为两卟啉之间的醚键断裂后的产物及其进一步加氢、N-甲基化和O-甲基化产物。结论华卟啉钠在大鼠体内以Ⅰ相代谢产物为主。
Objective To investigate the main metabolites and metabolic pathways of sodium humectin in rats by intragastric administration. Methods The plasma, urine and feces of rats after intragastric administration of sodium porphyrin were collected and the metabolites in urine, feces and plasma of rats were studied by Thermo Q Exactive Orbitrap quadrupole-orbit-trap high resolution mass spectrometry Obtained the accurate molecular weight and secondary fragment map, deduced the metabolites in urine, feces and plasma of SD rats, and mapped the metabolic pathway of sodium porphyrin in SD rats. Results A total of 10 metabolites were detected in the urine, feces and plasma samples from rats, mainly the products of the ether bond cleavage between two porphyrins and their further hydrogenation, N-methylation and O-methylation product. Conclusion Sodium humectin in rats with phase Ⅰ metabolites.
