用免疫组化方法,对大鼠暂时性全脑缺血诱导的c-fos原癌基因蛋白(Fos)在海马表达的分布,强度和时间过程等特征进行了观察.结果表明:缺血40min再循环后2h,首先在海马齿状回转折部的颗粒细胞和下托诱导出Fos表达,5~8h达到高峰,8h后锐减并逐步消失.CA4和CA3区的Fos表达在缺血再循环后3h出现,5h达到高峰,持续至24h消失.而在CA2和CA1区,Fos表达最弱,最晚(5h才出现),持续32~48h后消失.Fos在海马表达的强弱和时间发展过程依次为齿状回、下托→CA4、CA3→CA2、CA1区.这种特征性表达可能与介导缺血损伤后强啡肽、神经生长因子等晚期基因的表达有关.
The distribution, intensity and time course of c-fos proto-oncogene protein (Fos) expression induced by transient global cerebral ischemia in rat hippocampus were observed by immunohistochemistry.The results showed that: At 2h after circulation, Fos expression was induced in the granulosa cells and the hypothalamus of the dentate gyrus at hippocampus, and peaked at 5-8h, then dropped sharply and gradually disappeared after 8h.The expression of Fos in CA4 and CA3 was significantly increased after ischemia-reperfusion 3h, peaked at 5h and disappeared in 24h, while in CA2 and CA1 regions, Fos expression was the weakest (5h) and disappeared after 32 ~ 48h.The expression of Fos in hippocampus and time course Followed by the dentate gyrus, under the care → CA4, CA3 → CA2, CA1 area.The expression may be related to the expression of late genes such as dynorphin and nerve growth factor after ischemic injury.
