通过分析金针菇(Flammulina velutipes)子实体原基期、伸长期、成熟期的转录组数据,得到一个差异表达基因fv-msf1。对其结构、序列特征及表达情况进行分析,结果显示该基因全长2709 bp,包含13个外显子,12个内含子,开放阅读框长1770 bp,编码589个氨基酸。结构域分析表明该基因编码的蛋白含主要协助转运蛋白超家族(major facilitator superfamily,MFS)保守结构域。通过实时荧光定量PCR(real-time quantitative fluorescence PCR,q-PCR)对子实体不同发育阶段以及菌盖不同发育时期的表达量进行分析,结果显示该基因在金针菇伸长期的菌盖(尤其是0.3~0.9cm)中高表达,由此推断该基因在金针菇菌盖发育中发挥一定作用。
A differentially expressed gene fv-msf1 was obtained by analyzing the transcriptome data of protamine, elongation and maturation of Flammulina velutipes. The results showed that the full length of this gene was 2709 bp, including 13 exons and 12 introns. The open reading frame was 1770 bp in length and encoded 589 amino acids. Domain analysis indicated that the protein encoded by this gene mainly contributed to the conserved domains of major facilitator superfamily (MFS). Real-time quantitative fluorescence PCR (q-PCR) was used to analyze the different developmental stages of the fruiting body and the expression of different stages of the growth of cap, the results showed that the gene in the elongated mushroom cap (especially 0.3 ~ 0.9cm), suggesting that this gene plays a role in the growth of Flammulina velutipes cap.
