目的:通过检测趋化因子CCR2,CXCL9在早、中、晚期口腔黏膜下纤维性变(OSF)组织中基因及蛋白表达的改变,进一步探讨OSF的发病机制。方法:收集早、中、晚期OSF组织,RT-PCR检测趋化因子CCR2,CXCL9基因mRNA的表达,Western blot检测组织中趋化因子CCR2,CXCL9蛋白的表达。结果:趋化因子CCR2,CXCL9基因mR-NA及蛋白在早、中、晚期OSF组织中与对照组相比均呈现表达下调,在早、中、晚期病变组织中表达依次递减。在早期OSF病变组织中,CCR2,CXCL9基因mRNA较健康对照组分别上调406%和2051%,蛋白表达分别升高385%和1506%;在中期OSF病变组织中,CCR2,CXCL9基因mRNA较健康对照组分别上调369%和1901%,蛋白表达分别升高310%和1419%;在晚期OSF病变组织中,CCR2,CXCL9基因mRNA较健康对照组分别上调357%和1760%,蛋白表达分别升高309%和1401%。结论:早、中、晚OSF各期组织中趋化因子CCR2,CXCL9基因mRNA和蛋白水平表达均异常升高。此类因子的异常表达可能与OSF的发病机制相关。
OBJECTIVE: To investigate the pathogenesis of OSF by detecting the expression of chemokines CCR2 and CXCL9 in early, middle and late stages of oral submucous fibrosis (OSF). Methods: Early, middle and late OSF tissues were collected. RT-PCR was used to detect mRNA expression of chemokines CCR2 and CXCL9. Western blot was used to detect the expression of chemokines CCR2 and CXCL9. Results: The expressions of CCR2 and CXCL9 chemokines mR-NA and protein in the early, middle and late OSF tissues were down-regulated compared with the control group, and decreased in the early, middle and late stages of the disease. In the early stage of OSF, CCR2 and CXCL9 mRNAs were up-regulated by 406% and 2051%, respectively, and their protein expressions were increased by 385% and 1506%, respectively. In the medium OSF lesions, the mRNA expressions of CCR2 and CXCL9 were significantly higher than those in healthy controls Group were up-regulated by 369% and 1901% respectively, the protein expression increased by 310% and 1419% respectively. In the OSF group, the mRNA expressions of CCR2 and CXCL9 were up-regulated by 357% and 1760% % And 1401%. Conclusion: The mRNA and protein expression of chemokines CCR2 and CXCL9 in early, middle and late stages of OSF are abnormally elevated. The abnormal expression of these factors may be related to the pathogenesis of OSF.
