目的探讨抑癌基因Ⅰ型辅酶脱氢酶1α亚复合物13(NDUFA13)抑制结肠癌生长的机制。方法以人结肠正常组织为对照,用实时荧光定量PCR及免疫组织化学方法检测NDUFA13及miR-7在60例人结肠癌中的表达;体外细胞模型研究NDUFA13基因对结肠癌细胞株增殖水平的影响。结果与人结肠正常组织比较,NDUFA13及miR-7在人结肠癌表达显著下调(P<0.01),两者的表达呈正相关;在人结肠癌细胞株SW480及HCT-116中高表达NDUFA13能够显著抑制结肠癌细胞的增殖。结论抑癌基因NDUFA13在人结肠癌中通过促进miR-7的成熟而抑制肿瘤的发生发展。
Objective To investigate the mechanism of tumor suppressor gene type Ⅰ coenzyme dehydrogenase 1α subunit 13 (NDUFA13) inhibiting the growth of colon cancer. Methods Human colon normal tissue was used as control. The expression of NDUFA13 and miR-7 in 60 human colon carcinomas was detected by real-time fluorescence quantitative PCR and immunohistochemistry. The effects of NDUFA13 gene on the proliferation of colon cancer cell lines were studied in vitro . Results The expression of NDUFA13 and miR-7 in human colon cancer was significantly down-regulated compared with that in human normal colon (P <0.01), and the expression of NDUFA13 and miR-7 was positively correlated with that of normal human colon. NDUFA13 was highly expressed in human colon cancer cell lines SW480 and HCT-116 Colon cancer cell proliferation. Conclusion The tumor suppressor gene NDUFA13 inhibits tumor development in human colon cancer by promoting the maturation of miR-7.
