目的:研究鲨鱼软骨多糖对血管内皮细胞生长、新生血管形成及小鼠实体瘤生长的影响作用。方法:以MTT法测定鲨鱼软骨多糖体外对内皮细胞增殖的影响;用鸡胚绒毛尿囊膜实验测定对血管生成的影响;建立荷瘤小鼠,并观察该提取物对小鼠抑制瘤的影响。结果:鲨鱼软骨多糖能显著抑制血管内皮细胞的生长,MTT法抑制率为(45.615±0.084)%;并可抑制鸡胚绒毛尿囊膜新生血管的形成,PBS对照组血管数目为18.750根,鲨鱼软骨多糖组血管数目为(7.750±1.545)根,差异有统计学意义,P<0.05。鲨鱼软骨多糖能抑制荷Lewis小鼠肿瘤生长,鲨鱼软骨多糖组肿瘤抑制率与生理盐水组相比差异有统计学意义,P<0.01;鲨鱼软骨多糖+环磷酰胺组则明显高于环磷酰胺组,差异有统计学意义,P<0.05。结论:鲨鱼软骨多糖可以抑制内皮细胞生长及新生血管的形成,进而发挥抗肿瘤作用。
Objective: To study the effect of shark cartilage polysaccharide on the growth of vascular endothelial cells, neovascularization and growth of solid tumors in mice. METHODS: The effect of shark cartilage polysaccharide on the proliferation of endothelial cells was measured by MTT assay; the effect of angiogenesis was determined by chicken embryo chorioallantoic membrane test; tumor-bearing mice were established and the effect of the extract on mouse tumor inhibition was observed. . RESULTS: Shark cartilage polysaccharide could significantly inhibit the growth of vascular endothelial cells, and the inhibition rate was (45.615±0.084)% by MTT assay. It also inhibited the formation of neovasculature of chicken embryo chorioallantoic membrane. The number of vessels in PBS control group was 18.750, sharks The number of vessels in the cartilage polysaccharide group was (7.750±1.545) roots, and the difference was statistically significant (P<0.05). Shark cartilage polysaccharide can inhibit the growth of Lewis mice. The tumor inhibition rate of shark cartilage polysaccharide group was significantly different from that of saline group (P<0.01). The shark cartilage polysaccharide and cyclophosphamide group was significantly higher than that of cyclophosphamide. Group, the difference was statistically significant, P <0.05. Conclusion: Shark cartilage polysaccharide can inhibit the growth of endothelial cells and the formation of new blood vessels, and then play an anti-tumor effect.
