目的观察谷氧还蛋白1(glutaredoxin1,Grx1)在高糖诱导的血管内皮细胞凋亡中的保护作用。方法在高糖环境中培养人脐静脉内皮细胞,制备人脐静脉内皮细胞损伤模型,细胞分为正常对照组、损伤组、Grx1预保护组。倒置显微镜下观察细胞形态、(MTT比色法)检测细胞活力以初步观察Grx1对细胞损伤的保护作用;利用Hoechst33258染色法荧光显微镜观察细胞凋亡的形态学变化;采用Annexin V-FITC/PI双染法,流式细胞仪检测Grx1对内皮细胞凋亡的影响。结果光镜观察结果显示,Grx1预保护组与损伤组比较细胞状态明显改善;MTT法显示高糖环境下,Grx1可减轻高糖引起的细胞活性降低;Ho-echst33258染色实验显示,Grx1可明显改善高糖所致的内皮细胞变形、皱缩等损伤表现。流式细胞术检测显示当Grx1存在时,细胞凋亡率明显下降。结论Grx1对高糖诱导的血管内皮细胞凋亡具有明显的保护作用。
Objective To observe the protective effect of glutaredoxin1 (Grx1) on apoptosis induced by high glucose in vascular endothelial cells. Methods Human umbilical vein endothelial cells (HUVECs) were cultured in high glucose environment to prepare human umbilical vein endothelial cell injury models. The cells were divided into normal control group, injury group and Grx1 pre-protection group. Cell morphology was observed under inverted microscope. Cell viability was detected by MTT assay to observe the protective effect of Grx1 on cell injury. Hoechst33258 staining was used to observe the morphological changes of apoptotic cells. Annexin V-FITC / The effect of Grx1 on the apoptosis of endothelial cells was detected by flow cytometry. Results The results of light microscopy showed that the cell morphology was significantly improved in Grx1-preprotected and injured groups. MTT assay showed that Grx1 could reduce the cell viability induced by high glucose under high glucose conditions. The Ho-echst33258 staining showed that Grx1 could be significantly improved High glucose caused by endothelial cell deformation, shrinkage and other damage performance. Flow cytometry showed that when Grx1 was present, apoptosis rate was significantly decreased. Conclusion Grx1 has a significant protective effect on the apoptosis of vascular endothelial cells induced by high glucose.
