目的 探讨雌、雄激素对前列腺中胰岛素样生长因子Ⅰ (IGF Ⅰ )基因表达的影响。 方法 采用斑点杂交结合图像分析技术 ,观察药物去势和未药物去势BPH患者前列腺组织以及正常组、去势组、外源性雄激素组、雌激素组大鼠前列腺组织中IGF ⅠmRNA表达情况。 结果 人体内睾酮水平降低后 ,前列腺中IGF Ⅰ基因表达明显减少 ;正常大鼠前列腺中IGF ⅠmRNA表达量是去势大鼠的 2 .6倍 ,给予外源性睾酮后两组IGF ⅠmRNA表达量变化不大 ;雌激素组大鼠前列腺中IGF ⅠmRNA表达量明显高于正常组 ,平均表达量为正常组的 1.8倍。 结论 雌、雄激素可以调节前列腺中IGF ⅠmRNA表达量 ,IGF ⅠmRNA表达改变可能与前列腺体积变化相关。
Objective To investigate the effect of estrogen and androgen on the gene expression of insulin-like growth factor Ⅰ (IGF Ⅰ) in prostate. Methods Dot blot hybridization and image analysis techniques were used to observe the expression of IGF Ⅰ mRNA in prostatic tissue of normal and castrated rats, exogenous androgen and estrogen groups in castrated and non-castrated BPH patients. Results After the testosterone level in the human body was reduced, the expression of IGF Ⅰ gene in the prostate decreased significantly. The IGF Ⅰ mRNA expression in the prostate of normal rats was 2.6 times that of the ovariectomized rats. The expression of IGF Ⅰ mRNA in the two groups after administration of exogenous testosterone In the estrogen group, the expression of IGF-ⅠmRNA in the prostate was significantly higher than that in the normal group, and the average expression level was 1.8 times that of the normal group. Conclusions Estrogen and androgen can regulate the expression of IGF Ⅰ mRNA in prostate, and the change of IGF Ⅰ mRNA may be related to the volume change of prostate.
