目的:优选大孔吸附树脂法纯化合欢花总黄酮的工艺参数。方法:以合欢花总黄酮含量为指标,通过单因素试验考察树脂型号、上样液质量浓度、洗脱剂种类及用量、除杂溶媒种类及用量等对大孔树脂纯化工艺的影响。结果:采用AB-8型大孔树脂,优选的纯化工艺为上样液质量浓度0.2 g·mL-1,上样量2 BV,用4 BV水洗除杂,4 BV 10%乙醇洗脱,5 BV 50%乙醇洗脱,收集50%乙醇洗脱液,总黄酮纯度达34.62%。结论:优选的工艺条件可较好地分离、纯化合欢花总黄酮。
Objective: To optimize the process parameters of purification of total flavonoids of Albizia by macroporous adsorption resin method. Methods: According to the content of total flavonoids of Acacia flowers, the effects of resin type, the concentration of sample solution, the type and amount of eluent, the type and amount of miscellaneous solvents on the purification of macroporous resin were investigated by single factor test. Results: Using AB-8 macroporous resin, the optimum purification process was as follows: the mass of sample solution was 0.2 g · mL-1, the sample volume was 2 BV, washed with 4 BV water, mixed with 4 BV 10% ethanol, 5 BV 50% ethanol elution, collecting 50% ethanol eluent, total flavonoids purity of 34.62%. Conclusion: The optimal conditions of the process can be better separation and purification of the total flavonoids of Acacia flowers.
