目的:观察调脂积冲剂对高脂饮食诱导的脂肪肝大鼠的防治作用及对肝脏PPAR-γmRNA表达的影响,探讨其可能作用机制。方法:将SD大鼠49只随机分成空白组、模型组、易善复组、调脂积组。上午,正常组给予普通饲料喂养,模型组、易复善组和调脂积组给予高脂乳剂;下午,两个治疗组分别给予对应药物,空白组和模型组给予生活饮用水灌胃。8周末处死实验大鼠。测定肝脏总胆固醇(TC)和甘油三脂(TG)的含量;HE染色观察肝组织病理变化;采用RT-PCR检测大鼠肝脏PPAR-γmRNA的表达。结果:肝组织出现明显的脂肪变和炎性侵润;与模型组相比,调脂积组TG明显降低;模型组大鼠肝组织PPAR-γmRNA表达减少,调脂积组则显著升高。结论:调脂积冲剂能增加肝脏PPAR-γmRNA的表达。
AIM: To observe the preventive and therapeutic effects of Tiaozhi Impinger on fatty liver in rats induced by high-fat diet and its effect on the expression of PPAR-γmRNA in liver, and to explore its possible mechanism. Methods: 49 Sprague-Dawley rats were randomly divided into blank group, model group, Yishenfu group and lipid regulating group. In the morning, the normal group was fed with normal feed, the model group, Yi Fu Shan group and the lipid-lowering group were given high-fat emulsion. In the afternoon, the two treatment groups were given the corresponding drugs, the blank group and the model group were given drinking water. Experimental rats were sacrificed at 8 weeks. The contents of total cholesterol (TC) and triglyceride (TG) in the liver were measured. The pathological changes of liver were observed by HE staining. The expression of PPAR-γmRNA was detected by RT-PCR. Results: Compared with the model group, the TG of the lipid-lowering group was significantly lower than that of the model group. The expression of PPAR-γmRNA in the liver tissue of the model group was decreased and the lipid-lipid group was significantly increased. Conclusion: Tiaozhi capsule can increase the expression of liver PPAR-γmRNA.
