目的:探讨EB病毒(Epstein-Barr virus,EBV)潜伏膜蛋白1(Latent membrane protein 1,LMP1)基因特异性的脱氧核梅(DNAzyme)对伯基特淋巴瘤细胞P3HR-1的增殖和凋亡的影响。方法:设计合成针对EBV-LMP1基因的DNAzyme(DZ1),在P3HR-1细胞上观察其对细胞增殖活性、细胞周期和细胞凋亡的影响。结果:脱氧核酶DZ1作用于P3HR-1细胞后,可显著抑制EBV-LMP1基因的表达,并呈剂量依赖性。将脱氧核酶DZ1抑制LMP1的表达,进而下调抗凋亡蛋白Bc1-2的表达,诱导细胞色素c释放,从而诱导细胞凋亡。结论:在P3HR-1细胞中,靶向LMP1可能成为潜在的基因治疗的分子靶,靶向LMP1的脱氧核酶可能成为EB病毒相关肿瘤基因治疗实验研究的新的工具。
Objective: To investigate the proliferation and apoptosis of Burkitt’s lymphoma cell line P3HR-1 induced by Epstein-Barr virus (EBV) -specific Latent membrane protein 1 (LMP1) gene. Impact. Methods: DNAzyme (DZ1) against EBV-LMP1 gene was designed and synthesized. The effect of DNAzymes on the cell proliferation, cell cycle and apoptosis was observed on P3HR-1 cells. Results: The DNAzyme DZ1 inhibited the expression of EBV-LMP1 gene in a dose-dependent manner in P3HR-1 cells. DNAzyme DZ1 inhibits the expression of LMP1, which in turn down-regulates the expression of anti-apoptotic protein Bc1-2 and induces the release of cytochrome c to induce apoptosis. CONCLUSIONS: Targeting LMP1 may be a potential molecular target for gene therapy in P3HR-1 cells. LMP1 targeting deoxyribozymes may be a new tool for the experimental study of EBV-related tumor gene therapy.
