目的 研究慢性粒细胞性白血病 (CML)细胞冻融抗原 (CLA)负载的树突状细胞 (DC)对特异性抗白血病T细胞的作用。方法 将CML患者外周血单个核细胞 (PBMNC)来源的DC在体外用CLA负载 ,再与CML患者的经细胞因子诱导的杀伤细胞 (CIK)共同培养 ,应用乳酸脱氢酶 (LDH)释放法观察其对自身CML细胞杀伤活性 (A组 ) ,与未负载的DC +CIK(B组 )、CIK(C组 )及CIK +CLA(D组 )进行比较。结果 效靶比为 10∶1条件下 ,A、B、C、D 4组的杀伤活性分别为 (6 3 6 9± 8 35 ) %、(4 5 0 2± 5 49) %、(2 7 2 8± 4 6 4) %、(2 9 6 3± 5 71) %。A组比B组杀伤活性强 (P <0 0 1) ;C组与D组比无显著性差异。结论 CLA负载的DC可进一步提高DC介导的特异性抗白血病T细胞对CML细胞的杀伤活性
Objective To study the effects of CML-loaded dendritic cells (DCs) on specific anti-leukemic T cells in chronic myelogenous leukemia (CML) cells. Methods DCs derived from peripheral blood mononuclear cells (PBMNCs) from patients with CML were loaded with CLA in vitro and co-cultured with cytokine-induced killer cells (CIK) from CML patients. Lactate dehydrogenase (LDH) release was observed. The cytotoxicity against self CML cells (group A) was compared with unloaded DC + CIK (group B), CIK (group C), and CIK + CLA (group D). Results The killing activity of A, B, C, and D 4 groups was (6 3 6 9± 8 35 )%, (4 5 0 2± 5 49) %, and (2 7) when the effect-to-target ratio was 10:1. 2 8± 4 6 4) %, (2 9 6 3± 5 71) %. The killing activity of group A was higher than that of group B (P < 0 01); there was no significant difference between group C and group D. Conclusion CLA-loaded DCs can further enhance DC-mediated killing activity of specific anti-leukemic T cells against CML cells.
